    摘要:

    为提高食品中致病菌快速检测的准确性,设计了 一种基于三磷酸腺苷(ATP,adenosine 5′-triphosphate)生物发光反应动力学数据拟 合以及光学校准的食品致病菌检测系统。采用与ATP生物 发光波长相同的LED模拟生物发光,对检测样品的 光吸收及散射进行 校准,从而减小食品颜色及浊度的干扰。采用光电倍增管(PMT)连续记录ATP生物 发光强度数据,基于生物发光反应动力学方程自动对发光强度数据拟合,减小系统噪声并提 高仪表的测量 范围。研究了基于LED的光学校准算法,基于ATP生物发光动力学研究了数据拟合算法 ,进而设计实现了致病菌检测系统。对金黄色葡萄球菌样品的检测结果表明,本文系 统与 传统培养计数 方法相关性达到0.98,检测致病菌范围为102~108 CFU/mL,对单一样品测量的精密度 为5.05%。本文系统设计为生物发光 在食品安全检测领域的实际应用提供了新的思路,具有重要实际意义。

    Abstract:

    In order to improve the reliability of food pathogen detecti on,a system based on (adenosine 5′-triphosphate (ATP) bioluminescence reactio n kinetics data fitting and optic calibration is developed. An LED with the same wavelength as ATP bioluminescence is used to simulate the light absorption and scattering of the food samples.By this method,the food col or and turbidity interference are reduced.A photomultiplier tube (PMT) is used to record ATP intensity data con tinuously.Based on bioluminescence reaction kinetics equation,the bioluminescenc e intensity data is fitted to reduce the system noise and improve the measuring range.Optic calibration algorithm b ased on LED is presented,and data fitting algorithm based on ATP bioluminescence kinetics is studied.A pathogen detection system based on bioluminescence and optic calibration is studied and dev eloped in this work.S.aureus(ATCC26001) samples are detected and the results sho w that the system has a good linearity with the traditional culture counting method with correlation coeffici ent of 0.98.The detection range of pathogens is from 102CFU/mL to 108CFU/m L.The reproducibility of the luminometer is tested and the coefficient variation is within 5.05%.This work may provide a new strategy for ATP bioluminescence used in food hygiene and safety.
